How Amitriptyline and Pregabalin work Amitriptyline and Pregabalin is a combination of two medications: Amitriptyline and Pregabalin. Amitriptyline is a tricyclic antidepressant that increases the levels of chemical messengers (serotonin and noradrenaline) that inhibit the transmission of pain signals in the brain. Together, they alleviate neuropathic pain, which is pain resulting from damaged nerves. Pregabalin is an alpha-2 delta ligand that reduces pain by modulating the calcium channel activity of nerve cells.
