It works by interfering with the synthesis of ergosterol, a crucial component of the fungal cell membrane. Disrupting this membrane prevents the fungus from growing and kills its cells.

This lotion is prescribed for the topical treatment of various skin mycoses (fungal infections) that affect the body's surface. This includes common conditions like Tinea pedis (Athlete's foot), Tinea cruris (Jock itch), and Tinea corporis (Ringworm). Because it is a lotion, it spreads easily over larger or hairy areas of the skin, providing effective, convenient treatment.

NULL is a highly effective dermatological preparation formulated as a lotion for the localised treatment of superficial fungal skin infections. The active ingredient, Amorolfine, is distinct from other common topical antifungals (like azoles) because of its unique mechanism of action.

Amorolfine works through two primary effects:

1. Fungistatic Action: It inhibits the production of ergosterol, a vital substance for maintaining the structure and function of the fungal cell membrane.
2. Fungicidal Action: By altering the cell membrane structure, it leads to the accumulation of abnormal sterols, resulting in the death of the fungal cell.

The concentration in the lotion base is ideal for treating Tinea infections on the body and between the toes. The lotion format provides several advantages:

• Ease of Application: It spreads easily across both hairy and non-hairy skin areas.
• Rapid Absorption: The formulation allows the active ingredient to penetrate the upper layers of the skin, reaching the site of infection where the fungi reside.
• Targeted Treatment: It ensures a high, localised concentration of the antifungal agent directly at the site of the infection, minimising systemic absorption and potential side effects.

Successful treatment with NULL requires consistent daily application for the full prescribed duration, which often extends beyond the disappearance of visible symptoms to ensure complete eradication of the fungus and prevention of recurrence.

NULL (Amorolfine Lotion) is indicated for the topical treatment of dermatophytoses and other superficial fungal infections of the skin:

Tinea pedis (Athlete's foot):

Fungal infection typically between the toes or on the soles of the feet.

Tinea cruris (Jock itch):

Fungal infection affecting the groin area.

Tinea corporis (Ringworm):

Fungal infection appearing as circular, itchy, red patches on the body.

Pityriasis Versicolor:

A fungal infection causing small, discoloured patches on the trunk or arms.

Cutaneous Candidiasis:

Fungal infections of the skin caused by Candida yeast.

Common Side Effects (Usually Mild and Localised):

• Local Irritation: Transient stinging, burning, or redness (erythema) immediately after application.
• Pruritus: Mild itching or mild skin scaling/peeling at the treatment site.
• Dryness of Skin.

Rare Side Effects (Stop treatment and consult a doctor if you experience):

• Contact Dermatitis: A delayed, persistent, or worsening rash/inflammation indicating a local allergic reaction to the product or its ingredients.
• Severe Allergic Reaction: Although extremely rare, seek urgent medical help if you experience signs of a widespread rash, swelling of the face, or difficulty breathing.

NULL is for external topical use only. Do not swallow it and avoid contact with eyes, ears, and mucous membranes (mouth, nose, vagina).

• Clean and Dry: Thoroughly wash and dry the infected area of skin before each application.
• Application: Apply a thin layer of the lotion to the infected skin area and the immediate surrounding skin, usually once or twice daily (morning and/or evening), or as directed by your doctor.
• Rub In Gently: Gently rub the lotion into the skin until absorbed.
• Duration: It is crucial to continue using the lotion for the full duration prescribed by your doctor, even after the visible symptoms (such as redness or itching) have cleared. Treatment duration varies depending on the type and extent of the infection, often lasting several weeks.
• Hand Hygiene: Wash your hands thoroughly after applying the lotion, unless the treated area is your hands.

The active ingredient, Amorolfine, is a potent morpholine antifungal that interferes with the fungal cell's ability to maintain its structure and function.

1. Ergosterol Synthesis Inhibition: Amorolfine acts by inhibiting two key enzymes involved in the synthesis pathway of ergosterol, the cholesterol equivalent found in fungal cell membranes.

3. Fungicidal Effect: The resulting membrane damage prevents the fungus from growing and replicating (fungistatic effect) and ultimately leads to the breakdown and death of the fungal cell (fungicidal effect). This dual action provides effective clearance of the infection.

• Prevent Spread: For Athlete's foot (Tinea pedis), wear clean socks every day and dry your feet thoroughly, especially between the toes, after washing.
• Continue Treatment: The absence of symptoms does not mean the infection is cured. The treatment must be completed as prescribed to ensure all fungal elements are destroyed and to prevent the infection from returning.
• Clothing: To aid recovery and prevent spread, wear loose-fitting, breathable cotton underwear (for Tinea cruris).
• Avoid Contamination: Do not allow the lotion to come into contact with the eyes, nose, or mouth. If accidental contact occurs, rinse thoroughly with water.
• Nail vs. Skin: Note that this 0.25% lotion is for skin infections. Nail infections require a separate, higher-concentration Amorolfine lacquer (nail paint). Do not use this lotion for nail fungus.

• Store the lotion bottle at room temperature, typically below 25 °C or 30 °C, as specified on the packaging.
• Keep the bottle tightly closed when not in use.
• Protect the lotion from excessive heat and direct light.
• Ensure the medication is stored out of the reach and sight of children.

There are no known interactions between NULL topical lotion and food or beverages.

No known significant drug-drug interactions exist because systemic absorption of Amorolfine from the skin is negligible.

• Other Topical Agents: Avoid applying other topical creams, ointments, or lotions to the same infected area at the same time as NULL, as they may interfere with the absorption or effectiveness of Amorolfine.

Amorolfine lotion is generally safe for use in patients with other diseases, as systemic absorption is minimal. However, it should be used with caution:

• Hypersensitivity: Avoid use if there is a known allergy or hypersensitivity to Amorolfine or any other ingredient in the lotion base.
• Immunosuppression: In patients with severe immune suppression, healing may be slower, and complete eradication of the infection may require longer treatment.

Apply a thin layer of the lotion to the affected skin area once or twice daily, as directed by the physician.

The total duration of treatment is specific to the infection:

• Tinea pedis and Tinea corporis may require 2 to 3 weeks of continuous application.
• Pityriasis Versicolor may require 2 to 3 weeks.
• Cutaneous Candidiasis may require up to 4 weeks.

An overdose from the topical application of NULL is extremely unlikely due to minimal absorption into the bloodstream.

• Action: If a large amount of the lotion is accidentally swallowed, seek immediate medical attention or contact emergency services.

If you forget to apply the lotion at the scheduled time:

• Apply it as soon as you remember.
• If it is nearly time for your next scheduled dose, skip the missed one and continue with your regular schedule.
• Do not apply a double amount of lotion to make up for a missed dose. Consistency in application is more important than making up for a missed single dose.
